TMEM100 Regulates Neuropathic Pain by Reducing the Expression of Inflammatory Factors

There is no effective treatment for peripheral nerve injury-induced chronic neuropathic pain (NP), which profoundly impacts the quality of life those affected. Transmembraneprotein100 (TMEM100) considered to be a regulatory protein and expressed in dorsal root ganglion (DRG) rats. However, mechanism regulation expression TMEM100 following various injuries are unclear. In this study, we constructed two models injury: tibial injury (TNI) constriction (CCI). This study found that Paw Withdrawal Mechanical Threshold (PWMT) Withdraw Thermal Latency (PWTL) rats decreased significantly, DRG groups also significantly. Furthermore, decrease CCI group was more obvious than TNI group. significant statistical significance ( P > 0.05 ). We an adeno-associated virus 6 (AAV6) vector expressing recombinant fluorescent injected it into sciatic (SN) models: TNI. PWMT PWTL were significantly increased groups, along with spinal cord DRG. It inhibited activation microglia, astrocytes, several inflammatory mediators (TNF- α, IL-1 β, IL-6). summary, results suggested might promising molecular strategy NP, its anti-inflammatory effects play important role relief.
